The Ready-to-Assay GPR54/KiSS1 Receptor Frozen Cells are a simple, thaw-and-assay cell format for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This assay assesses ligand based activation of KISS1R GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses KISS1R alongside endogenous high levels of a promiscuous G-protein, coupling the receptor to the calcium signaling pathway. Upon activation of KISS1R, the resulting calcium activity can be fluorescently assayed. Ready-to-assay kits consist of two vials of the stably modified clonal cell line in a convenient thaw-and-use format. Ki-67 antigen is a nuclear, non-histone protein that is present in all stages of the cell cycle except G0. This characteristic makes Ki-67 an excellent marker for proliferating cells and is commonly used as one of the prognostic factors in cancer studies. A correlation has been demonstrated between Ki-67 index and the histo-pathological grade of neoplasms. Assessment of Ki-67 expression in renal and ureter tumors shows a correlation between tumor proliferation and disease progression, thus making it possible to differentiate high-risk patients. Ki-67 expression may also prove to be important for distinguishing between malignant and benign peripheral nerve sheath tumors. Ki-67 labeling index has been shown to be a prognostic marker in a number of neoplasms including grade II astrocytoma, oligodendroglioma, colon carcinoma, and breast carcinoma. Measure arrestin signaling kinetics in real time with the live cell, fluorescent Borealis arrestin - LPA1 receptor assay. Add the unmodified LPA1 receptor and Borealis sensor to cells and measure robust signals the next day on an automated plate reader or fluorescence imaging system. This assay is genetically encoded and packaged in BacMam for efficient transduction in most cell types - including uniform and near 100% efficiency in HEK293 cells as well as expression in many primary or iPSC-derived cells. The kit includes the arrestin sensor BacMam, LPA1R BacMam, 500mM sodium butyrate in H2O, and a positive control receptor and agonist (AT1R BacMam and 5mM Angiotensin II in H2O). Borealis kits for several other receptors are also available, and larger-sized kits are available on request. Other Montana Molecular tools including the cADDis cAMP and DAG sensors can be used alongside this assay.
